Stem cell technology could help harness patients' own immune cells to fight disease

Jan. 3, 2013 ? The human body contains immune cells programmed to fight cancer and viral infections, but they often have short lifespans and are not numerous enough to overcome attacks by particularly aggressive malignancies or invasions. Now researchers reporting in two separate papers in the January 4th issue of the Cell Press journal Cell Stem Cell used stem cell technology to successfully regenerate patients' immune cells, creating large numbers that were long-lived and could recognize their specified targets: HIV-infected cells in one case and cancer cells in the other.

The findings could help in the development of strategies to rejuvenate patients' exhausted immune responses.

The techniques the groups employed involved using known factors to revert mature immune T cells into induced pluripotent stem cells (iPSCs), which can differentiate into virtually any of the body's different cell types. The researchers then expanded these iPSCs and later coaxed them to redifferentiate back into T cells. Importantly, the newly made T cells were "rejuvenated" with increased growth potential and lifespan, while retaining their original ability to target cancer and HIV-infected cells. These findings suggest that manipulating T cells using iPSC techniques could be useful for future development of more effective immune therapies.

In one study, investigators used T cells from an HIV-infected patient. The redifferentiated cells they generated had an unlimited lifespan and contained long telomeres, or caps, on the ends of their chromosomes, which protect cells from aging. This is significant because normal aging of T cells limits their expansion, making them inefficient as therapies. "The system we established provides 'young and active' T cells for adoptive immunotherapy against viral infection or cancers," says senior author Dr. Hiromitsu Nakauchi, of the University of Tokyo.

The other research team focused on T cells from a patient with malignant melanoma. The redifferentiated cells they created recognized the protein MART-1, which is commonly expressed on melanoma tumors. "The next step we are going to do is examine whether these regenerated T cells can selectively kill tumor cells but not other healthy tissues. If such cells are developed, these cells might be directly applied to patients," says senior author Dr. Hiroshi Kawamoto, of the RIKEN Research Center for Allergy and Immunology. "This could be realized in the not-so-distant future."

The 3 Best eZine Formats | Online success with SWELEX's business ...

eZine publishers today use any one or more of the 3 most commonly used eZine formats which I am about to describe to you in this article.

Ultimately, you can decide which format is the best choice for you, though each winning format has its pros and cons.

The text eZine is the most commonly published. The advantage of this format is that other than writing, you do not require any special skill to use a Word or Notepad program.

While the HTML eZine format requires a certain degree of HTML skills on your part, you can add more sophisticated features to your eZine issues, making them more appealing to your subscriber thus increases your readership value, something that text eZines do not have.

You can decorate your eZine format, change your fonts, include pictures, and more. However, the drawback often faced by HTML eZines is that they often get trapped into spam filters before they reach their subscribers? inboxes.

The third and least used among the 3 formats is the PDF eZine. Publishing your eZine in PDF format can consume a lot of time and effort on your part but often make up in quality readership

Due to the commitment, PDF eZines are usually published on a monthly basis. The great part, though, is that you can put in your affiliate links in your PDF eZine issue and allow your subscribers to pass the eZine issue around.

Given the choices, however, you do not have to necessarily choose strictly one eZine format, as some eZine publishers today do publish in more than one format.

96% Argo

All Critics (248) | Top Critics (43) | Fresh (237) | Rotten (11)

'Argo' is one of the best movies of the year.

Argo has that solid, kick-the-tires feel of those studio films from the 70s that were about something but also entertained. Only it's as laugh outright amusing as it is sobering.

The movieland satire is laid on thick, but it's also deadly accurate. Schlock has never seemed so patriotic, and Arkin and Goodman have rarely been so good.

Argo is a rollicking yarn, easily the most cohesive and technically accomplished of Affleck's three films so far, but a part of me wishes the director hadn't cast himself in the lead role.

If nothing else, it proves that every so often, the CIA can pull something off - and that yes, Canadians are just about the nicest people on the planet.

The film is a whopper of a tale, one designed for Oscar nominations, Best Picture and Best Director among them.

A strong yet uneven effort...

We are invested in this rescue mission not simply because these are fellow Americans in peril, but also because Argo takes the time to make each person behind enemy lines unique and authentic.

An edge-of-your-seat thriller not to be forgotten at Oscar time!

'Argo' only clocks in at a two-hour running time and tells a much better story than the films that over-stuffed audiences with nearly three-hours.

Second billing to unabashed jingoism

This may be the most geek-friendly movie that never did the rounds at Comic-Con.

When Tony (Ben Affleck) lands in Tehran and trains the frightened Americans to play their parts as a Canadian film crew, Argo compares the ruses run by governments and movie studios.

The whole thing is paced like a whip, and the third act may literally be the most suspenseful piece of film I've ever seen - and yes, I have seen Rear Window. Recommended without any reservation for every and any cinemagoer.

A terrific supporting cast, especially the non A-list actors, make the dramatic license taken justified and redeem Affleck for miscasting himself in the hero's role.

A brilliant thriller that's based on a historical incident, 'Argo' is one of the best films of 2012.

A wonderfully lively and engrossing movie ...

An enthralling and deliciously entertaining movie -- and a deserved triumph for its star and director.

The details make for a rip-roaring story, which Affleck invests with old-school directorial ?lan.

Ben Affleck leaps on to the A-list of directors with this relentlessly entertaining thriller, combining comedy and nerve-jangling suspense to maximum effect.

If you're a nailbiter, bring gloves, because the suspense will see you reach your knuckles.

Surprisingly enjoyable, given that you may have little prior commitment to this story and only mildly fancy Affleck.

Affleck skilfully turns the screw of suspense, gaining added traction from an extremely convincing recreation of the setting and period.

Ben Affleck's movie tells an amazing but true story set against the context of the Iranian hostage crisis.

Talent borrows and genius steals, but Affleck does something in between: he mimics.

Pak assumes presidency of UN Security Council | Pakistan Today ...

  • PM will be in New York to chair January 21 open debate on peacekeeping

UNITED NATIONS?-?Pakistan on Tuesday assumed this month?s presidency of the UN Security Council and will hold special sessions on peacekeeping and counterterrorism.

?During its presidency, Pakistan will help Council members to forge consensus and bridge differences. Ours will be an effective presidency,? Pakistan?s Permanent Representative to the UN Masood Khan said.

Pakistan began its two-year term as non-permanent member of the 15-nation Security Council last year and will be the president of the UN body for January, the first time that Pakistan has assumed the rotational office of the UNSC during its present term.

Prime Minister Raja Pervez Ashraf would be in New York to chair the January 21 open debate on ?UN Peacekeeping: a Multidimensional Approach?, which would take a comprehensive view of UN peacekeeping.

Foreign Minister Hina Rabbani Khar would chair a ministerial level session on January 15 on counterterrorism.

Khan said the debate would provide an opportunity to the council members and the general UN membership to have a ?holistic view of the continuing threats and challenges posed by international terrorism and the best ways of formulating and implementing coherent and comprehensive responses to this menace?.

?We have put the difficult issue of counterterrorism on the agenda of the council for a special debate. Terrorism can only be rooted out through a comprehensive approach of deterrence, dialogue and development,? Pakistan?s UN envoy added.

Previously, Pakistan has been elected to the Security Council six times.

The UN Security Council has five veto-wielding permanent members ? the United States, Britain, France, Russia and China ? and 10 nations that are elected as non-permanent members without veto powers.

Big loser of new laws: Sex-offender Santas

By Tracy Connor, NBC News

?

Rich Pedroncelli / AP

California hunters will no longer be able to use dogs to hunt bobcats and bears. Here, Josh Brones, president of the California Houndsmen for Conservation, walks his hunting dogs, Dollar, left, Sequoia, center and Tanner right, near his home in Wilton.

With every new year comes a hodgepodge of fresh local laws ? many serious, some silly, and others so obvious it makes you wonder what took legislators so long.

The National Conference of State Legislatures has compiled a year-end list of some of the laws that go on the books Jan. 1.

Among the winners: California bobcats and Illinois residents who overshare on Facebook. Losers include randy police officers and sex offenders with Santa suits.

In Maryland, same-sex couples will get the right to marry. In California, clergy who oppose gay unions don?t have to perform them.

California also outlawed hound-hunting of bobcats and bears after deciding it isn?t a fair fight.

"There is nothing sporting in shooting an exhausted bear clinging to a tree limb or a cornered?bobcat," state Sen. Ted Lieu said when the bill he authored was passed.

Also now prohibited in California: law enforcement officers having sex with anyone in custody, including prisoners who have been arrested but not yet booked.

California and Illinois have both gone to bat for social networkers who want to keep snarky status updates and bikini photos under wraps, barring employers from forcing job applicants or workers to hand over passwords for Facebook and Twitter.

New Year kicks off on Christmas Island, begins rolling west
Beer now considered alcohol, not food, in Russia

Illinois is also looking out for military re-enactors, expanding an exemption from gun laws to include weapons with barrels less than 16 inches long.

Other new Illinois laws: a ban on shark fins and a law prohibiting sex offenders from handing out Halloween candy or dressing up like Santa or the Easter bunny.

Florida is putting the brakes on swamp buggy drivers who want to tool around on state roadways; they can?t, unless local law explicitly allows it.

But the Sunshine State has some good news for drivers who flash their headlights to let oncoming motorists know that police have set a speed trap up ahead. They can no longer be ticketed for it, come the first of the year.

Leadership in the Medical Field: Series Part 1?What It Is & Why It's ...

Today we have the first post in a three-part series from our regular?contributor, Lindsey Harper Mac. Lindsey is a professional writer living in the Indianapolis area. She specializes in writing guest posts on social media and education. Currently, Lindsey is completing work on her master?s degree. You can find links to some of her earlier posts at the end of the article.

Leadership in the medical and healthcare field is a critically important subject for healthcare personnel, those interested in the field and the public to examine at some length. Leadership determines the quality of healthcare provided to patients even during periods of relative stasis. Thus, the topic needs to be provided to all healthcare provider students, from those currently completing a medical assistant program to postdoctoral physician fellows. Further, the authority provided by effective leadership helps maintain measured calm through the upcoming periods of change scheduled to take place in the US through 2020 according to the Affordable Care Act of 2010. Through this series of three articles, we plan to examine what?s meant by the term ?medical leadership,? why it?s so important to both professionals in the field and patients, what personal attributes are required to be an effective leader and the means by which effective leaders work with other healthcare providers.

Effective medical leadership is more than the ability to bark out instructions and orders for subordinates to follow and complete. Rather, as Matt Green and Lynne Gell indicate in their recent article for BMJ Career?s website, ?Effective Medical Leadership for Consultants: Personal Qualities and Working with Others,? the term is best defined through a theoretical framework that includes the many qualities required of the term and the interactive ways in which it?s demonstrated. As O*Net Online indicates?for a representative healthcare position, a registered nurse (RN)?leadership in the healthcare field also includes aspects of ?strategic planning, resource allocation, human resources modeling?. and coordination of people and resources.?

The Medical Leadership Competency Framework

In order to illustrate the qualities, types of qualities and the means of demonstrating leadership, the National Health Service (NHS) Institute for Innovation & Improvement and the Academy of Medical Royal Colleges together developed the Medical Leadership Competency Framework ?annotated diagram above. This figure allows physicians and other healthcare workers to visually review the different aspects of medical leadership and self-assess themselves as to areas that might require improvement or training. The diagram is in the shape of a circle to represent the holistic ideal of quality of medicine. Within the circle?s core is the ultimate goal, ?Delivering the Service.? The center of the circle opens to the outside via five separate lanes that effectively divide the outer aspect of the framework?s circle into five wedge-like areas that represent the ways in which leadership is demonstrated:

  • Demonstrating personal qualities.
  • Working with others.
  • Managing services.
  • Improving services.
  • Setting direction.

Each of these wedge-like areas is further comprised of four primary characteristics indicating how leadership in that particular area is best demonstrated. In Part II and Part III of this article series, we?ll explore what the four primary characteristics are for two of the areas, ?demonstrating personal qualities? and ?working with others.?

Why is Effective Medical Leadership Deemed ?Critical??

As Green and Gel summarized in their paper cited earlier, the connection between effective medical leadership and good medical care is ?well documented? and the ?key to delivery high quality care and a positive experience for patients.? In part 2 of this series, we?ll discuss further the specific personal qualities that make a good leader in the medical field.
